Sinai Health Systems – Toronto, Ontario, Canada
The Ontario Familial Colorectal Cancer Registry (OFCCR) site recruited 2186 CRC-affected case probands via Cancer Care Ontario, and 287 CRC-affected and 60 unaffected probands via genetic clinics across Ontario. CRC cases diagnosed between the ages of 20 and 74 years (Phase-I) and under the age of 50 (Phase-II) were recruited with greater emphasis on families with multiple cases of CRC. In addition, 4784 primarily 1st– and at-risk 2nd-degree relatives ascertained through the family proband and 1948 CRC-unaffected control participants ascertained via the Ministry of Finance were recruited.
Cleveland Clinic – Cleveland, Ohio, USA
The Cleveland Clinic CCFR site recruited 193 CRC case probands and 68 unaffected probands via Cleveland Clinic. Families with HNPCC or HNPCC-like history—defined as a family with 3 or more CRC cases (or 2 CRC & 1 uterine, or 2 CRC plus polyps), where 1 was diagnosed under the age of 50 years, and 1 was a 1st-degree relative to the other 2 in successive generations—were recruited. In addition, 266 CRC-affected and 543 unaffected 1st-, 2nd– and 3rd-degree relatives ascertained through the family proband were recruited.
The University of Melbourne – Melbourne, Victoria, Australia
The Australasian Colorectal Cancer Family Registry (ACCFR) site recruited 1296 CRC cases and 204 unaffected probands via Victoria Cancer Registry and genetic clinics in Adelaide, Brisbane, Melbourne, Perth, Sydney, and Auckland, New Zealand. CRC cases diagnosed under the age of 60 years (Phase-I) and under the age of 50 (Phase-II) were recruited with greater emphasis on families meeting Amsterdam I/II criteria, with strong history of CRC or HNPCC, or with a known germline mismatch repair (MMR) gene mutation or with an MMR deficient CRC tumor. In addition, 9758 1st-, 2nd– and 3rd-degree relatives were recruited. Unaffected participants ascertained via Federal electoral roll (n=270), their 1st-degree relatives (n=179) and 449 CRC-unaffected spouses were recruited to serve as controls.
University of Hawai’i – Honolulu, Hawai’i, USA
The Hawai’i CCFR site recruited 529 CRC cases via The Surveillance, Epidemiology, and End Results (SEER) Registry for the state of Hawai’i. CRC cases diagnosed between the ages of 18 and 84 years (Phase-I), under the age of 50 years (all races) and 18 – 84 years of Japanese American ancestry (Phase-II) were recruited. In addition, 1021 primarily 1st-degree relatives and 180 CRC-unaffected case spouse controls participants ascertained via the case were recruited.
Mayo Clinic – Phoenix, Arizona, USA
The Mayo Clinic CCFR site recruited 1255 CRC cases and 23 unaffected probands via Minnesota Cancer Surveillance System (MCSS), the Mayo Clinic, Rochester and the North Central Cancer Treatment Group. CRC cases diagnosed in the state of Minnesota between the ages of 18 and 74 years (Phase-I) and under the age of 50 or with a strong family history of CRC (Phase-II) were recruited. In addition, 2988 primarily 1st-degree and 2nd-degree relatives in the cancer side of the family and 254 CRC-unaffected spouse control participants ascertained via the case were recruited.
Fred Hutch Cancer Center – Seattle, Washington, USA
The Seattle Colon Cancer Family Registry (SCCFR) site recruited 2354 CRC cases via the Puget Sound SEER Registry and 20 CRC-affected and 26 unaffected probands via the Seattle Cancer Care Alliance and the Hereditary Cancer Clinic at Providence/Swedish Medical Center. CRC cases diagnosed 20 and 74 years from King, Pierce and Snohomish counties (Phase-I) and under the age of 50 from all 13 western Washington counties (Phase-II), or presenting to clinic with a known germline mutation in MMR or MutYH gene, or meeting the Familial Colorectal Cancer Type X family criteria were recruited. In addition, 5896 primarily 1st– and CRC-affected 2nd– and 3rd-degree case relatives ascertained through the family proband and 2058 CRC-unaffected controls ascertained via the Washington State Department of Licensing were recruited.
University of California – San Francisco, California, USA
The Cancer Prevention Institute of California (CPIC) CCFR site (located at the University of California, San Francisco (UCSF)) recruited 176 CRC cases via Northern California and Greater Bay SEER Registries during Phase II. CRC cases diagnosed between the ages of 18 and 74 years of African American and Japanese American ancestry from northern California and the greater Bay area were recruited. In addition, 131 1st– and CRC-affected 2nd– and 3rd-degree case relatives and 42 CRC-unaffected case spouse controls participants ascertained via the case were recruited.
